Mitochondrial proteases in human diseases
نویسندگان
چکیده
Mitochondria contain more than 1000 different proteins, including several proteolytic enzymes. These mitochondrial proteases form a complex system that performs limited and terminal proteolysis to build the proteome, maintain, control its functions or degrade proteins peptides. During protein biogenesis, presequence cleave targeting signals obtain mature functional proteins. Processing by also exerts regulatory role in modulation of quality enzymes misfolded, aged, superfluous Depending on their substrates, defects can affect majority proteome only single protein. Consequently, mutations have been linked human diseases. This review gives an overview components machinery highlights pathological consequences dysfunctional processing turnover.

mitochondrial dysfunction is one of the main causative factors in a wide variety of complications such as neurodegenerative disorders, ischemia/reperfusion, aging process, and septic shock. decrease in respiratory complex activity, increase in free radical production, increase in mitochondrial synthase activity, increase in nitric oxide production, and impair in electron transport system and/or...
متن کاملMultifunctional Mitochondrial AAA Proteases
Mitochondria perform numerous functions necessary for the survival of eukaryotic cells. These activities are coordinated by a diverse complement of proteins encoded in both the nuclear and mitochondrial genomes that must be properly organized and maintained. Misregulation of mitochondrial proteostasis impairs organellar function and can result in the development of severe human diseases. ATP-dr...
متن کاملComparative mitochondrial proteomics: perspective in human diseases
Mitochondria are the most complex and the most important organelles of eukaryotic cells, which are involved in many cellular processes, including energy metabolism, apoptosis, and aging. And mitochondria have been identified as the "hot spot" by researchers for exploring relevant associated dysfunctions in many fields. The emergence of comparative proteomics enables us to have a close look at t...
متن کاملUnsolved issues related to human mitochondrial diseases.
Human mitochondrial diseases, defined as the diseases due to a mitochondrial oxidative phosphorylation defect, represent a large group of very diverse diseases with respect to phenotype and genetic causes. They present with many unsolved issues, the comprehensive analysis of which is beyond the scope of this review. We here essentially focus on the mechanisms underlying the diversity of targete...
متن کاملMitochondrial Dynamics in Mitochondrial Diseases
Mitochondria are very versatile organelles in continuous fusion and fission processes in response to various cellular signals. Mitochondrial dynamics, including mitochondrial fission/fusion, movements and turnover, are essential for the mitochondrial network quality control. Alterations in mitochondrial dynamics can cause neuropathies such as Charcot-Marie-Tooth disease in which mitochondrial f...
